cDNA microarray Analysis Identifies Symptom-Associated Chemokine Genes in Behcet's Disease Mouse Model

Behcet's disease(BD), a chronic inflammatory disease, is characterized by intraocular inflammation, oral ulcers, genital ulcers, skin lesions and additional accompanying changes. A developing aspect of BD is reached until eye symptoms are present from early mucocutaneous symptoms via arthritic and neurological symptoms, or systemic symptoms are present at the same time. BD eye symptom in particular represents a systemic results from a serious sequela. To understand these serious changes of symptoms, this study tried to observe differences in gene expression that correlated to BD mucocutaneous symptoms turning into BD eye symptoms. We used the BD mouse model infected with herpes simplex virus composed of typical BD mucocutaneous symptomatic mice without ocular symptoms(BD skin), BD ocular symptomatic mice with mucocutaneous symptoms(BD eye), and asymptomatic mice despite HSV inoculation(BD inactive). Using Japanese criteria with minor modification, We classified the mice by one major and one minor symptom. Using peripheral blood mononuclear cells from each group, a cDNA microarray was performed. microarray chips were composed of 7,744 mouse genes. The BD skin group was compared with the BD eye group, and BD inactive group. The genes expressed differently and we selected genes of related to inflammation out of the 7,744 genes, those confirmed by reverse transcriptase-polymerase chain reaction(RT-PCR). The results were reconfirmed via protein expressions coupled with immunohistochemistry and western blot analysis. CCL28 was newly discovered in PBMC of BD skin. In the skin region, CCL28 was observed in sabaceous glands and corneous tissue. CCL28 was higher in BD skin than BD inactive and BD eye. In the receptor of CCL28, CCR10 was higher in BD skin and BD eye than BD inactive regions of skin inflammation. CCR3 was higher in PBMC of BD eye than BD inactive and BD skin. However, CCR7 was lower in BD skin relative to BD inactive and BD eye. The expression patterns of CCL19, a ligand of CCR7, was similar to CCR7 in lymph nodes. The expression patterns of CCL21, a ligand of CCR7, was expressed similarly among groups within the germinal center and lymphatic vessel of the lymph node. These results suggest CCR7, CCL28, their receptor and ligands are related to BD symptom patterns in our mouse model.
