The Role of PKC-δ in Hypoxia-induced Cerebrovascular Endothelial cell death.

dc:descriptionIn various cell types, PKC(protein kinase C)-δ has been found to be an important mediator during hypoxia-induced cell death. However, the role of PKC-δ in cerebrovascular endothelial cells is unknown. Therefore, we investigated the role of PKC-δ during hypoxia-induced cell death using bEnd.3 cell, a cerebrovascular endothelial cell line. The activity of PKC-δ increased remarkably after 1 hour of hypoxic injury, and the increase was maintained until 4 hours of hypoxia. To investigate the role of PKC-δ during hypoxia in terms of cell death, we measured the amount of released LDH (lactate dehydrogenase) and trypan blue positive cells during hypoxia after treatment with rottlerin, PKC-δ inhibitor. Hypoxia-induced LDH release was significantly increased by the treatment with rottlerin. We examined the specific blockade of PKC-δ activation by rottlerin after 1 hour of hypoxia in bEnd.3 cells. When pcDNA-PKC δ-WT was transfected into bEnd.3 cells, LDH release and trypan blue positive cells were attenuated during hypoxia. From these results, it is suggested that hypoxia causes an activation of PKC-δ, rendering bEnd.3 cells resistant to hypoxia-induced cell death.
