DNA -Binding Proteins That Facilitate Integration of Transmissible DNA Elements in Bacteroides Thetaiotaomicron
Abstract
dc:descriptionI implemented a biochemical approach utilizing heparin column, size-exclusion and DNA-affinity chromatography, 2-D gel electrophoresis and mass fingerprinting to identify proteins that bound the attachment sites of CTnDOT (attDOT ) and NBU1 (attNI), and stimulated integration in vitro for both DNA elements. The proteins isolated and characterized include BHP, a non-specific DNA-binding protein that is most likely involved in nucleoid organization, BHP2, a homolog to BHP that probably functions in a similar capacity, and BT 1847, a specific DNA-binding protein that is a member of the AraC/Xy1S transcription regulator family. Preliminary data suggest that another protein, BT1271, works in conjunction with BT1847 to confer DNA-binding and specificity. BT1271 is annotated in the genome as the 30S ribosomal subunit S16.
