Finally, soluble high-affinity TCRs are promising reagents for the detection and diagnosis of specific virus or tumor antigens (peptide-MHC) on the cell surface. In addition, the TCRs would provide a useful probe for tracking the processing and presentation of specific antigens. Chapter 6 discusses the development of soluble single-chain TCRs with enhanced properties, such as increased avidity and signal amplification, to detect antigen. The results show that tetrameric and multi-fluorophore labeled forms of the high-affinity 2C TCR called m67 were capable of detecting low levels of specific peptide-MHC on the surface of both tumor cells and when the antigen was cross-presented on stromal cells in a solid tumor.
