Absorption, Distribution and Excretion of Oxytetracycline and Tetracycline in Female Beagles

The absorption, distribution and excretion of oxytetracycline and tetracycline were studied in female beagles following intra-arterial and oral drug administration. Concentrations of drug in body fluids and selected tissues were determined by high performance liquid chromatography utilizing an internal standard. The average biologic half-life of oxytetracycline was 6.4 hours, whereas that of tetracycline was 13.7 hours following intra-arterial administration (10 mg/kg body weight). The half-life of tetracycline was decreased to 8.2 hours when a lower dose (5 mg/kg body weight) was administered. Oxytetracycline and tetracycline were rapidly but incompletely absorbed when administered to canines per os. Both antibiotics had similar absorption (< 0.5 hr) and elimination (4.8-6.6 hrs) half-lives. Comparisons of the recovery of both drugs in urine following intra-arterial and oral drug administration revealed that tetracycline had a higher bioavailability (35.9%) than that determined for oxytetracycline (10.7%). Following intra-arterial administration of oxytetracycline (10 mg/kg body weight) the highest concentration of drug was present in the kidney, followed by the liver, heart, muscle, and lung in declining order of concentration. A positive correlation was demonstrated between coexistent mean plasma and tissue concentrations for the tissues examined.
