Enhance heme biosynthesis in Saccharomyces cerevisiae

This research delves into the enhancement of heme production in Saccharomyces cerevisiae through Adaptive Laboratory Evolution (ALE) and Atmospheric and Room Temperature Plasma (ARTP) mutagenesis, complemented by strategic nutrient supplementation. Despite the theoretical promise of ALE and ARTP to induce beneficial mutations for augmented heme biosynthesis, the results highlighted the difficulties in modifying metabolic pathways for intended enhancements. Additionally, the supplementation of ferric ions and glycine, aimed at increasing the availability of precursors for heme biosynthesis, did not yield a significant increase in heme production, as evidenced by quantitative measurements of heme and its direct precursor, protoporphyrin IX (PpIX). The challenges encountered in the high-throughput screening for mutants with enhanced heme production underscored the need for more refined detection methods to identify and quantify metabolic improvements accurately. This research contributes to the understanding of the limitations and potentials of using evolutionary and mutagenesis techniques in yeast strain development, emphasizing the need for an integrated approach that combines genetic, biochemical, and analytical strategies to advance microbial production of valuable metabolites.
