Verifying the Deletion of Growth Hormone Receptor Using a Quantitative Polymerase Chain Reaction at the mRNA Level in Tissue-Specific GHR-/- Mice

dc:descriptionGrowth Hormone (GH) inhibits insulin action, and high level GH can result insulin resistance and diabetes in both mice and humans. GH receptor (GHR) gene disrupted mice (GHR-/- mice) have been genetically modified to lack the GHR and GH action. GHR-/- mice are extremely insulin sensitive and have an extended life span. The contribution of individual tissues to the phenotype of GHR-/- mice is not known but of extreme interest in the aging field. Three tissues likely to contribute to the improved insulin sensitivity are liver, adipose tissues, and muscle, as these are insulin and GH sensitive. To examine the individual contributions of these tissues to the overall insulin sensitivity and longevity in GHR-/-mice, three tissue-specific GHR-/- mice were created using a Cre-Lox system. This thesis represents the efforts to verify the deletion of GHR at the mRNA level only in the specific tissues of these tissue-specific gene-disrupted mouse lines. Collectively, these data show the GHR mRNA levels are reduced only in the tissues targeted with no significant reduction in any other tissue. Thus, we have successfully created mouse lines with GHR deletion specifically in the liver, muscle, or fat tissue. These mice are now ready for more thorough phenotypic analyses.
