Influence of Oxygenated Perfluorocarbon (oxyPFC) on survival and free radical production of Ischemic Cardiomyocytes

dc:description.abstractIn the late 1980’s, several animal studies showed that oxyPFC given intravenously or via intracoronary route have the potential to reduce the size of myocardial infarctions. The exact mechanism by which this reduction occurred remained unclear until today. The subject of this experimental work was to elucidate the pathways of interaction of oxyPFC with hypoxic cardiomyocytes. The first mechanism we elucidated in this study was the effect of oxyPFC on mitochondrial respiration in a hypoxic state of the cardiomyocyte, and the second was to find out which role oxygen free radicals, especially superoxide, play after administration of oxyPFC. Several cellular biological methods were used to analyse mitochondrial respiration under the influence of oxyPFC. The results show that myocardial viability and ATP-production is increased after application of small doses of oxyPFC. The results further indicated that superoxide is up-regulated in an early phase after oxyPFC application, and down-regulated 6 hours after oxyPFC application. This experimental study clarified some of the physiological mechanisms by which oxyPFC reduces the infarct size. Our results suggest that oxyPFC may be useful as an adjunct therapy in myocardial infarction to increase survival of cardiomyocytes.
