Identification of binding protein with kinesin18B

Microtubules play an important role in intracellular transport of vesicles and organelles in higher eukaryotic cells. This transport depends on the activity of motor proteins that interact with microtubules. Most kinesins move cargoes outwards toward the cytoplasmic membrane. Members of the kinesin family have been implicated in vesicle and organelle transport, spindle and chromosome motility during meiosis and mitosis, microtubule destabilization, and flagellar and ciliary function. In the present study, we identified a protein that interacts with kinesin18B using a yeast two-hybrid system, GST pull-down assay, immunoprecipitation. This protein is Microtubule-associated protein RP/EB family member 1 containing a calponin homology(CH) domain at their N terminus and an EB1-like C-terminal motif at their C terminus : the functional importance of these domains has not been determined. Human EB1 family proteins, which include EB1, EBF3, and RP1, also associate with the tumor suppressor protein adenomatous polyposis coli(APC) and p150glued, a component of the dynactin complex. EB1 interacts with kinesin18B and has been localized to clusters near the distal ends of microtubules. Kinesin18B has been shown to move along microtubules and has been located at the plasma membrane and nuclear. The interaction between kinesin18B and EB1 may help to identify of function of kinesin18B.
