Einfluss der Inhalationsanästhetika Sevofluran und Desfluran auf die Ausbildung von Adhäsionsmolekülen auf Granulozyten in vitro

dc:descriptionVolatile anesthetics are reported to reduce ischemia-reperfusion injury. One suggested mechanism was the attenuated expression of CD11b on activated neutrophils after exposure to volatile anesthetics. However, neutrophil adhesion to vascular endothelium is a multistep process involving several selectins and b2- integrins. Therefore, we assessed wether sevoflurane and desflurane affect the activation of the selectins PSGL-1 (CD162) and L-selectin (CD62L) and the b2- integrins CD11a and CD11b. Whole blood was incubated for 60 minutes with 1 or 2 MAC (minimum alveolar anesthetic concentration) sevoflurane or desflurane. After incubation, neutrophils and monocytes were activated with N-formyl-methionyl-leucyl-phenylalanine (FMLP) or phorbol-12-myristate-13-acetate (PMA). Activation of adhesion molecules was evaluated by flow cytometry. The results of this study make clear that the volatile anesthetics have different effects on the adhesion molecules on neutrophil granulocytes (PMNs) and monocytes and that the effects are dependent on the concentration of the gases. Regarding the adhesion different results can be demonstrated. Partly an increased expression of the adhesion molecules can be showed, partly a decreased expression. This indicates an increased or reduced molecular adhesion.Further studies are required to identify the exact mechanisms of those effects to get a better understanding of reperfusion injury.
