Study of the Metabolic Effects of Water-alcoholic Extracts of Heartwoods with Traditional Use in Bulgaria // Проучване метаболитните ефекти на водно-алкохолни екстракти от дървесини с традиционно приложение в България

dc:description[EN] The interest in the therapeutic potential of Bulgarian medicinal plants in prophylaxis and treatment of various pathological conditions increased in the recent years. Many investigations are focused on the scientific explanations of the national traditions. Knowledge of the therapeutic effects of the arboreal species is based on the use of their fruits, leaves, flowers and bark. Polyphenol content and antioxidant activity of heartwoods traditionally used for coloring of high alcoholic beverages are not studied yet in Bulgaria. The toxicity and therapeutic potential of the mulberry (Morus nigra) heartwood ethanol infusion remains unknown. The aim of this study was to reveal the effect of Morus nigra heartwood ethanol infusion in models of t-ButOOH induced oxidative stress and LPS-induced inflammation in murine 3T3-L1 preadipocytes and J774A.1 macrophages. Based on the results obtained it could be suggested that mulberry heartwood ethanol infusion possesses antioxidant activity by inhibiting transcription levels of enzymes related to the endogenous antioxidant defense (GCL and GPx4) under conditions of oxidative stress and inflammation. Furthermore, scientific evidences were obtained about anti-inflammatory effect of the investigated extract demonstrated by decrease of gene expression of inflammatory cytokines and inflammatory enzymes such as NF-kB, TNF-a, IL-6, IL-lb, COX-2, iNOS, ICAM -1 and the increase of Sirt-1 mRNA levels for in LPS-stimulated macrophages. Based on the reported data we concluded that the Morus nigra heartwood infusion could be a potential source of active compounds with therapeutic potential in prevention and treatment of various chronic diseases.
