Estrogen Inducible Serpin, Proteinase Inhibitor 9, in Immunosurveillance and Apoptosis

The phytoestrogen genistein has attracted a great deal of recent interest. In MCF-7 cells, we find that 10 nM genistein induces PI-9 as effectively as saturating E2, and genistein treated MCF-7 cells are resistant to NK cell mediated cytotoxicity. In ERalphaHA cells, the dose response curves for induction of PI-9 by E2 and by genistein are nearly identical and inducing PI-9 with genistein blocks NK cell mediated killing. Analysis of MCF-7 tumors produced in a mouse xenograft model shows that genistein, soy-products including soy flour and NovasoyRTM induce PI-9 expression in vivo. Our data highlight the possibility that low concentrations of genistein might promote growth of pre-existing estrogen receptor-dependent breast cancer.
