Total Genomic Transcriptional Analysis of the Fatty Acid Degradation Regulon of Escherichia Coli

The second ORF, b2323 is the fabB gene which produces beta-ketoacyl-ACP synthase I. This is one of two long chain specific condensing enzymes in E. coli and is required for unsaturated fatty acid biosynthesis. The other enzyme necessary for producing unsaturated fatty acids is 3-hydroxy-decanoyl-ACP dehydrase, encoded by the fabA gene. This gene has previously been shown to be positively regulated by FadR. Double fabA(Ts), fadR mutants have a synthetic lethal phenotype, fabB(Ts), fadR strains were constructed and shown to behave in a similar fashion. Experiments involving chloramphenicol acetyl transferase fusions confirm that FadR positively regulates transcription of fabB as well.
