Proline Utilization in Salmonella Typhimurium: A Model for Regulation of Peripheral-Membrane Flavin Dehydrogenase-Lipid Interactions
Abstract
dc:descriptionWhen it is located in the cytoplasm, PutA exists as a soluble dimer that is available to bind put operator DNA. However, proline oxidation and available membrane lipid allow PutA to undergo a dimer-to-monomer transition that is required for membrane association. On the other hand, if membrane lipid is not readily available PutA remains in the soluble, dimeric conformation and represses transcription of the put operon. Thus, although both proline and membrane lipid are required for induction of the put operon, a quaternary transition appears critical to prevent overexpression toxicity by this peripheral membrane flavin dehydrogenase.
