Oxygen Regulated Genes in Salmonella Typhimurium
Abstract
dc:descriptionStudies on oxd-2 showed that it encodes a 42-kD protein which has acetate/propionate kinase activities. The cloned oxd-2 gene, under a foreign promoter, complemented an ackA (encoding an acetate kinase) mutation. Both Oxd-2 and AckA have a similar substrate range, and Oxd-2 has higher affinity for propionate than for acetate. Transcription of oxd-2 was induced to high levels during anaerobic growth in an amino acid rich medium. The highest level was found in cells grown anaerobically in a medium containing serine as a carbon source and supplemented with aspartate, leucine, and threonine. oxd-2 expression was affected by mutations in fnr, hip (IHF), crp, lrp, ackA or pta. NMR analysis of the cultures of a wildtype strain, grown anaerobically in a medium containing threonine, aspartate, leucine and casamino acids, showed that S. typhimurium produced propionate from threonine.
