Abstract
dc:descriptionUsing a similar strategy of library construction and screening, I also isolated the Vibrio harveyi acyl-ACP synthetase gene and showed that it encoded a 60.4 KDa protein that shared high homology with medium chain fatty acid-CoA ligases. I expressed the His-tagged protein at a high level in E. coli and purified it to homogeneity. To explore the physiological application of this enzyme in E. coli fatty acid metabolism, I constructed the first E. coli fabH (beta-ketoacyl-ACP synthetase III) bypass mutant in the presence of acyl-ACP synthetase and exogenous fatty acids.
