Negative Regulation of the accBC Operon in Escherichia Coli

Expression of the accB gene from B. subtilis complemented E. coli AccB activity, but did not cause negative regulation of the accBC genes. A chimeric protein consisting of the amino-terminal 44 amino acids of E. coli AccB fused to the carboxyl-terminal biotinylation domain of B. subtilis AccB complemented AccB activity and caused negative regulation of the accBC operon. Complementation of ACC activity using the malonyl-CoA synthase gene (matB) from Rhizobium trifolii allowed the deletion of the accA and accB genes. Transcriptional activity of the accBC operon in this strain shows an absence of growth-rate dependent regulation.
