Identification and Characterization of a Tolerant Response in Arabidopsis Thaliana to Tobacco Ringspot Virus and Cloning of a Single Locus That Confers Tolerance

dc:descriptionThe TTR1 locus was mapped using the progeny of crosses of Col-0 and Estland. In the initial mapping, PCR-based short sequence length polymorphism and cleaved amplified polymorphic sequence markers located across the A. thaliana genome were scored on 31 F$\sb3$ families to determine the chromosome location of TTR1. This study placed TTR1 between markers DFR and nga129 on the bottom arm of chromosome V, and further fine mapping using 446 F$\sb3$ families placed the locus 5.8 cM from DFR and 6.3 cM from nga129. Using informative F$\sb3$ families and restriction fragment length polymorphism markers, TTR1 was located in the interval between CIC2A12LE and yUP18A5LE, with genetic distances of 0.62 cM and 0.23 cM, respectively. No recombination was observed between RFLP probe yUP5F5LE and TTR1. CIC2A12LE, yUP18A5LE, and yUP5F5LE all hybridized with yeast artificial chromosome (YAC) clone CIC2A12, suggesting that this YAC clone contains the TTR1 locus. A cDNA clone, 120K3T7, also co-segregated with TTR1 in 430 F$\sb3$ families. Cosmid libraries of the YAC clone CIC2A12 (containing Col-0 TTR1 locus) and A. thaliana Estland genomic DNA (containing Estland ttr1 locus) were constructed.
