Enzymatic and Splicing Activity of Class I Leucyl -Trna Synthetases From Escherichia Coli and Saccharomyces Cerevisiae

Leucyl-tRNA synthetase from yeast mitochondria is also required for splicing of the mitochondrial bI4 and al4a introns. Splicing of these introns is dependent on coordination between ymLeuRS and bI4 maturase. An in vitro splicing assay has been established that is based on assembly of proteins into a competent ternary splicing complex. RNA deletion mutants have been quantitated for effects on complex assembly and splicing activity.
