Small Molecule Rhenium Carbonyl Complexes: Model Integrated-Design Technetium Estrogen Receptor Ligands for Tumor Imaging

It has been shown that inclusion of an acyl substituent on the cyclopentadiene ring lowers the pKa of the ring hydrogens sufficiently for labeling in an aqueous buffer. A divergent synthetic route to a pair of acyl-substituted cyclopentadienyl complexes, along with their alkyl analogs, has been developed. A modification of the cyclopentadienyl motif introduces a hydroxyl-substituted 2-pyridinylmethyl group onto the central ring. The inclusion of the pyridine moiety allows for displacement of one CO as the ligand wraps around the rhenium. The low electrophilicity of a beta-aryl-substituted-cyclopentenone has thus far been a complicating factor in the completion of these syntheses.
