Structure and Metabolism of Heparin and Heparan Sulfate
Abstract
dc:descriptionHeparin was selectively cleaved at N-sulfoglucosamine residues by treatment with nitrous acid at pH 1.5. The oligosaccharide products, primarily disaccharides and tetrasaccharides, were then end-labeled by reduction with sodium borotritide and purified to homogeneity using a combination of BioGel P-2 chromatography and strong anion exchange HPLC. The unknown components of both the disaccharide and tetrasaccharide fractions were identified using a combination of chemical and enzymatic steps designed to convert each oligosaccharide to compounds of known structure. Following structure elucidation, the purified compounds were used a standards for the development of several HPLC based procedures in which all of the known structures could be identified and quantified in any heparin sample of interest.
