Polymorphism of bovine MHC class II genes: Association with persistent lymphocytosis caused by bovine leukemia virus

Polymorphism of bovine major histocompatibility complex (MHC) class II DQA, DQB, and DRB genes was investigated by sequence comparisons between class II genes of cattle and other species. The association between BoLA-DRB3 alleles and persistent lymphocytosis (PL) caused by bovine leukemia virus infection was studied in detail. The relationship between PL and allelic amino acid sequences of BoLA-DRB3 exon 2 derived from three resistance- and four susceptibility-associated haplotypes was examined. Amino acids Glu-Arg (ER) at positions 70-71 were found to be present only in resistant haplotypes, whereas the motif Val-Asp-Thr-Tyr at positions 75-78 was commonly found in haplotypes associated with susceptibility to PL. The presence of the ER motif in resistance-associated DRB3 alleles was confirmed by a polymerase chain reaction (PCR)-based typing method in twenty-six unrelated cows, 13 of which were PL. A 783 bp BoLA-DQA cDNA clone 5) was identified by screening a bovine cDNA library. The α5 clone has an average nucleotide similarity of 91.6%, 86.0% and 84.2% with a BoLA-DQA genomic clone, SLA-DQA, and HLA-DQA, respectively. In contrast, similarity was 74.4% and 63.7% with bovine DYA and DRA, respectively. The polymorphism of exons 2 and 3 of BoLA-DQB cDNA was studied by reverse-transcription-PCR cloning and sequencing. Sequences characteristic of DQB1 and DQB2 genes were identified, demonstrating that both DQB genes are expressed. BoLA-controlled resistance and susceptibility to PL can be used as an animal model for studying the role of the MHC in retroviral diseases of humans and other animals.
