Label-free multi-modal multiphoton imaging for real-time point-of-procedure assessment of needle biopsies

Needle biopsy (NB) is known as a standard clinical procedure for cancer diagnosis in humans and companion animals. There has been a rising concern about the time cost of the conventional histopathological assessment of the NB specimens. Therefore, a real-time point-of-procedure assessment method of NB specimens is increasingly in demand. Label-free multimodal nonlinear optical imaging (NLOI) provides a powerful and non-invasive tool to study the microstructure and biochemical distributions in biological tissue, especially in tumor microenvironments. Continuous efforts have been devoted to the development and optimization of an intraoperative label-free multimodal NLOI system that integrates four NLOI modalities: two- and three-photon fluorescence, and second and third harmonic generation. Intraoperative NLOI of fresh NB specimens from tumor-bearing animals during veterinary surgeries demonstrated the potential of this system to enable real-time and point-of-procedure histological evaluation of the NB specimens for cancer diagnosis. Further investigations have been focused on using the label-free multimodal NLOI to monitor time-lapse changes in the tissue microenvironment of the fresh NB specimens sustained in a perfusion chamber.
