DESIGNING ANTI-PDL1 ANTIBODY CONJUGATED POLYMERIC COACERVATES AS A NOVEL “NANO-IMMUNOTHERAPY” AGAINST HEPATOCELLULAR CARCINOMA

Hepatocellular Carcinoma (HCC) accounts for a majority of primary liver cancer cases and is among the deadliest cancers in the world. With a shift in major risk factors from chronic viral hepatitis infections to that of metabolic diseases causing HCC, the number of HCC-related cases and deaths is expected to rise. While progress has been made in enhancing systemic therapies for HCC, resistance development and toxicity of these treatments are common. To circumvent these issues, a novel “nano-immunotherapy” to deliver encapsulated cationic polycarbonates for induction of immunogenic cell death and conjugated anti-PDL1 antibodies for immune checkpoint inhibition against murine hepatomas was thus designed. Several crucial insights have been gleaned which provide a roadmap for the improvement of future designs of nano-immunotherapies not only for a more robust and stable delivery of cationic polycarbonates but also for a more sustained and detectable localised and systemic immunity against murine hepatomas.
