The confocal scanning microscope has proven to be a powerful tool to produce high-quality three-dimensional (3D) imaging. However, the resolution of the confocal laser scanning microscope in the visible spectrum is restricted to be ~180 nm and ~500 nm in the lateral and axial direction respectively due to the optical diffraction. In this thesis, a novel technique, called the non-contact microsphere confocal scanning microscope, is proposed and developed. This setup can enhance the lateral resolution to 110 nm, the axial resolution to 100nm and image contrast to 71%, while retaining the key features of the conventional confocal microscope, such as no damage to samples, 3D in-vivo imaging and the ability to work in different environments.
