ΜΕΛΕΤΗ ΤΗΣ ΕΚΦΡΑΣΗΣ ΤΟΥ ΑΝΘΡΩΠΙΝΟΥ ΕΜΒΡΥΟΝΙΚΟΥ Ε-ΣΦΑΙΡΙΝΙΚΟΥ ΓΟΝΙΔΙΟΥ
Abstract
dc:descriptionTHE STUDY OF THE MOLECULAR MECHANISMS THAT TAKE PART IN THE REGULATION OF TRANSCRIPTION AND THE PROCESSING OF ITS PRODUCTS IS CENTRAL TO THE STUDY OF REGULATION OF GENE EXPRESSION. THE GLOBIN GENE SYSTEM IS ONE OF THE BEST STUDIED IN THIS DIRECTION. THE ANALYSIS OF TRANSCRIPTION AND RNA PROCESSING OF THE HUMAN EMBRYONIC E-GLOBIN GENE HAS BEEN STUDIED AND IS REPORTED IN THIS THESIS. WE ANALYZED TOTAL CELLULAR RNA FROM THE HUMAN HYBRID PUTKO CELL LINE AND FROM HELA CELLS TRANSIENTLY TRANSFECTED WITH THE E-GLOBIN GENE. FOR HELA CELL TRANSIENT EXPERIMENTS WE CONSTRUCTED A SERIES OF PLASMID VECTORS CONTAINING THE E-GLOBIN GENE AND, IN ALL POSSIBLE COMBINATIONS, THE SV40 ENHANCER SEQUENCES AND SV40 ORIGIN OFREPLICATION.
