ΜΕΛΕΤΗ ΤΩΝ ΦΥΣΙΚΩΝ ΠΛΑΣΜΙΔΙΩΝ ΤΟΥ ΣΤΕΛΕΧΟΥΣ ATCC 10988 ΤΟΥ ΑΙΘΑΝΟΛΟΠΑΡΑΓΩΓΟΥ ΒΑΚΤΗΡΙΟΥ ZYMOMONAS MOBILIS
Abstract
dc:descriptionFIVE NATIVE PLASMIDS OF Z. MOBILIS STRAIN ATCC 10988 HAVE BEEN ISOLATED, NAMELYPZMO2, PZMO3, PZMO5, PZMO6 WITH SIZE 1,70, 1,85, 2,70, 16,7, AND 31,8 KB RESPECTIVELY. RESTRICTION MAPPING (PARTIAL) OF ALL THESE PLASMIDS WAS FOLLOWED AND THEIR HOMOLOGY LEVELS WERE DETERMINED. SIMILARLY, THEIR HOMOLOGY AGAINST TOTAL GENOMIC DNA AND PLASMID DNA OF Z. MOBILIS STRAINS ATCC 10988, NCIB 11163 AND CP4 WAS EXAMINED. PLASMID PZMO3 WAS NONHOMOLOGUS EITHER TO CHROMOSOMAL DNA OR ANYOF THE OTHER PLASMIDS THESE STRAINS HARBOURED. A SERIES OF RECOMBINANT PLASMIDS WERE ESTABLISHED AND WERE USED TO CONFINE THE REGION OF PZMO3 WHICH CONTAINS SEQUENCES INVOLVED IN ITS STABILITY CONTROL. ONE OF THE SUBCLONED FRAGMENTS EXHIBITED STRONG INCOMPATIBILITY BETWEEN THE RESPECTIVE RECOMBINANT PLASMID AND NATIVE PZMO3. THIS HAS RESULTED INTO THE ESTABLISHMENT OF A VEGETATIVE CURING SYSTEM AGAINST THE NATIVE PLASMID (PZMO3), A METHOD WHICH PROVIDES, FOR THE FIRST TIME, AN ALTERNATIVE TOOL FOR STRAIN CONSTRUCTION VIA RECOMBINANT DNA IN Z. MOBILIS.
