dc:description.abstractGenetic alterations of the PTEN gene located on chromosome 10q23 have been found in different neoplasms. In squamous cell carcinoma of the head and neck (SCCHN) loss of heterozygosity (LOH) at 10q has been described to be associated with poor prognosis. Moreover, genetic instability of microsatellite repeat sequences (MI) seems to concur with PTEN mutation in endometrial and colorectal cancer. <br>We screened a panel of SCCHN for LOH at 10q23 using the microsatellite marker D10S541. In addition, instability of 11 microsatellite loci of 8 different chromosomes was determined. Also, all tumors were examined for p53 mutation. <br>All 36 patients assessed in this study had advanced disease and histologically confirmed complete resection of their tumors followed by standard radiotherapy of 60 Gy in 30 fractions. Cancer-specific survival and local-regional control were not significantly different for patients with or without LOH at D10S541. The same was found for p53 mutation and MI frequency. Only T stage, N stage and anemia were of prognostic value. <br>LOH at D10S541was observed in 7of 19 (37%) informative cases and correlated with MI. 80% of tumors with two or more loci with instability had LOH compared to 21% if instability was absent or limited to one locus only (p= 0.033). <br>In conclusion, our data confirm that LOH at 10q23 is a frequent event. Here we describe a significant correlation with MI positive tumors for the first time in SCCHN.
