Autoantibodies From Patients With Nonallergic Asthma Cause Cytotoxic Injury To Airway Epithelial Cells.

RATIONALE: Increased levels of IgG autoantibodies to airway epithelial cells in serum samples of nonallergic asthma compared to allergic asthma and healthy controls have been reported. And a possible involvement of autoimmune response against bronchial epithelial cell antigen in the pathogenesis of nonallergic asthma has been suggested. To evaluate a functional significance of autoimmune response to airway epithelial cells in the pathogenesis of nonallergic asthma, we examined autoantibody-mediated cytotoxicity to airway epithelial cells using purified IgG antibodies from patients with nonallergic asthma. METHODS: IgG antibodies were purified from patients with nonallergic asthma using by protein A column. IgG antibody-mediated cytotoxicity to cultured human airway epithelial cells (A549) was examined by microcytotoxicity assay using Terasaki tray. The cytotoxicities were expressed as % cell death. RESULTS: Purified IgG antibodies from patients with nonallergic asthma caused cytotoxic injury to airway epithelial cells in a dose-dependent manner. No significant cytotoxic injury to airway epithelial cells was observed by purified IgG antibodies from healthy controls. The cytotoxicities (% cell death) by IgG antibodies from nonallergic asthma (23.6 ± 5.0 %) were significantly higher than the cytotoxicities of IgG antibodies from healthy controls (9.6 ± 3.2 %) and allergic asthma (12.5 ± 2.9 %)(p<0.05). Incubation of IgG antibodies from patients with nonallergic asthma with airway epithelial cell proteins immobilized to solid phase (but not by bovine serum albumin) significantly inhibited the IgG antibody-mediated cytotoxicity to airway epithelial cells (p<0.05). CONCLUSION: This finding suggests a possible involvement of autoantibody-mediated cytotoxicity to airway epithelial cells in the pathogenesis of nonallergic asthma.
