TIS21 inhibits cell migration by downregulating expression of matrix metalloproteinases
Abstract
dc:descriptionTetradecanoyl phorbol acetate-inducible sequence 21 (TIS21) is the mouse homolog of rat pheochromocytoma cell 3 (PC3) and human B-cell translocation gene 2 (BTG2). TIS21 regulates cell cycle progression and proneural genes expression by acting as a transcription coregulator that enhances or inhibits the activity of transcription factors. And In human tissue, BTG2 has a critical role in cancer cell migration and invasion. The matrix metalloproteinases (MMPs) are zinc-dependent endopeptidases that are capable of degrading all kinds of extracellular matrix proteins, but also can process a number of bioactive molecules, and to perform key functions in tumor progression, and in other diseases. In this study, we report that TIS21 can decrease the expression at both mRNA and protein levels of MMPs in human breast cancer (MDA-MB-231) cells and human dermal fibroblasts (HDF) cells, as confirmed by zymography, RT-PCR. Moreover we demonstrate that TIS21 decreases transcription factor (c-Fos, PEA3) of MMPs. We are going to examine the regulatory mechanism of TIS21-mediated MMPs proteoytic activity.
