Entwicklung eines Systems für die kontrollierte Wirkstofffreisetzung aus biodegradierbarem Poly-D,L-Lactid zur Restenoseprävention im kardiovaskulären Bereich
Abstract
dc:descriptionThe biggest problem concerning the implantation of cardiovascular stents remains the restenosis.The CESP-procedure allows to integrate thermolabile substances in poly-D,L-lactid. By bioabsorption the agents are released in the body as a kind of a drug-delivery-system.The growth factor VEGF which is specific for endothelial cells shall reduce the proliferation of the smooth muscle cells by promoting the endothelisation of the stent. Colchicin causes by inhibitition of the SMC directly a reduction of the neointimale hyperplasia. In this work a satisfactory release and a good development of the effects of the drugs was detected at endothelial cells of human umbilical vein. The sterilisation of the foils by gamma-rays shows no influence to the potency of the substances.In the direct contact the settlement of the endothelial cells on the samples was insufficient This is mainly caused by the inhomogeneous, porous structure of the material. The scanning electron microscopy shows inhomogenity which has to be compensated by optimazing the method of production. The attempt of direct inhibition of the smooth muscle is thus more promising. The chemical prevention of restenosis by drug-delivery-system represents a good alternative to the brachytherapie.
