Etablierung eines Modells zur Isolation, Kultivierung und Stimulation kindlicher Herzmuskelzellen

Cardiomyocytes of children with congenital heart defects can be isolated from atrial biopsies and be kept in culture for a period of several weeks. Cultivated cardiomyocytes are capable of proliferation without loosing their typical properties, especially the ability to produce heart specific proteins such as myoglobine, troponine and alpha-sarcomeric-actine. Cultivated cardiomyocytes are able to produce pro-inflammatory cytokines after stimulation with LPS. There is a discrepancy between the detection of RNA of the respective cytokine and the amount that is, in fact, released into the interstitium. The reason for this may be an inhibition of the “posttranslational processing” by certain substances. In cultivated infant cardiomyocytes, hypothermia induces an attenuated production of proinflammatory cytokines, both with and without LPS stimulation. This result leads to the conclusion that hypothermia has protective effects for the heart, and effects due to cytokines such as heart insufficiency could probably be reduced. Our children model provides the opportunity to mirror the in-vivo-mechanisms in heart of children with congenital defects and has therefore many advantages compared to the conventional fetal or animal models. With the aid of our cultur model, there is the option to investigate further underlying pathomechanisms and to develop pharmacological and therapeutical strategies.
