The plasma concentration of apolipoprotein B (apoB)-containing lipoproteins is positively associated with several diseases, including atherosclerosis, type 2 diabetes and obesity. Hence, apoB may be a prime target for therapeutic intervention, particularly during its assembly. The molecular events associated with the initial formation of apoB-containing lipoproteins within the endoplasmic reticula of hepatic and intestinal cells are poorly understood. Earlier studies are consistent with a cotranslational lipid-desorption model that requires microsomal triglyceride transfer protein (MTP) and results in the formation of a small, neutral lipid core-containing emulsion particle. However, controversy exists as to the nature of the primordial lipoprotein and the requirement for MTP.
