Abstract
dc:descriptionBacteriophages, as obligate intracellular parasites of bacteria and archaea, represent the most diverse group of entities on Earth. They inhabit all ecological niches where their hosts are present and play a pivotal role in regulating bacterial populations and driving bacterial evolution. The complexity of interactions among bacteriophages, bacteria, and the host organism makes studying the intestinal microbiota particularly challenging due to the multitude of influencing factors. This study specifically examines the impact of a high-fat diet on these dynamic interactions. The advent of next-generation sequencing and advancements in metagenomic techniques have revolutionized the ability to assess gut microbiota composition without the necessity for microbial isolation and culture in the laboratory. Despite their transformative potential, these methods are not without limitations. Results can be confounded by background nucleic acids, rapid viral mutations, and the paucity of conserved sequences essential for accurate identification. Consequently, a significant proportion of sequences remain unclassified, often referred to as 'viral dark matter'. Laboratory isolation and cultivation of bacteriophages, however, provide an opportunity for in-depth study and facilitate further experimental applications. This approach also holds the potential for discovering novel bacteriophages, thus enriching viral databases with previously undescribed entities. This study aims to isolate aerobically culturable bacteriophages from mouse gut samples and evaluate the impact of a high-fat diet on their diversity. Bacteriophages and bacteria were isolated from mouse fecal samples provided by Dr. Aurelius Burokas’ team at the Institute of Biochemistry, Vilnius University. Bioinformatic analysis of genomic sequences, protein profiling of virions, and transmission electron microscopy (TEM) revealed the isolation of two genetically and morphologically distinct viruses: the siphovirus 50Rn infecting Enterococcus sp. strain MMBS 50 and the myovirus SuR infecting Serratia ureilytica DSM 16952. Both viruses are characteristic of the intestines of mice fed either a high-fat or a normal diet. Future studies are proposed to be conducted under anaerobic conditions to broaden the spectrum of bacteriophages and their hosts.
