Abstract
dc:descriptionMore and more people every day are getting diagnosed with cancer. It is very important to know exactly which biomolecules and other various compounds are in the cancerous tissue, how different environments are affecting these biotissues, and what the main mechanisms behind it are. This could help identify cancer even faster and more successfully. This study investigates the impact of various sample treatment methods on Surface-enhanced Raman scattering spectroscopy (SERS) of biotissue analysis. SERS is a very powerful tool to detect and help characterize biomolecules with high sensitivity and selectivity. However, this analysis method is easily influenced by various factors such as sample preparation, tissue preservation and fixation. Conditions and weight for the sample play a huge role in SERS analysis too. The main aim of this research is to systematically examine the impact of different samples (cancerous and non-cancerous) and how they are affected by changes in sample treatment, preparation, and preservation to understand the differences in SERS spectra of these samples. The ultimate goal is to identify which biomolecules are more sensitive and selective for the SERS method. This thesis begins with an analysis of the current scientific literature on Raman spectroscopy, SERS and the main differences between these methods. It also explores how nanoparticles, in addition to their shape and size, affect the SERS signal, and which biomolecules, groups, and compounds are most SERS selective and sensitive. In the experimental section, various biotissues (cancerous and non-cancerous) SERS spectra are examined over time. We also observe that the preparation of the sample shows different spectra of these biotissues. The storage conditions of the samples affect the SERS spectra depending on the solution in which the samples are placed. The results from this investigation shows that some Raman shifts peaks are specific to cancerous (483 cm-1, 667 cm-1, 1216 cm-1, 1586 cm-1, 1004 cm 1) and non-cancerous (1004 cm-1, 1325 cm-1) kidney tissue, independently of the treatment or the medium in which the tissue was stored. When deionized water is used for biosample storage, degradation products of biosamples are observed after a shorter period of time. When using formaldehyde, almost all Raman shifts of spectra correspond to the Raman shift bands of the formaldehyde Raman spectra. In general, this investigation of biosamples provides valuable insight into samples preparation, fixation and preservation, and their effects on SERS, the spectral profile, and selectivity of biomolecules.
