Salmonella spp., Listeria monocytogenes ir Yersinia enterocolitica are attributed to the most common foodborne pathogens, that can be detected in such products as poultry, pork, raw vegetables and fruits. Real time PCR is an accurate and rapid method for quantitative identification of foodborne pathogens in food matrix. In order to achieve maximum sensitivity of the method, two techniques were chosen: multiplication of the initial amount of pathogenic bacteria by cultivating chicken wing rinses and increase of the DNA amplification efficiency by using PCR additives (ethylene glycol and bovine serum albumin) in the reaction mixture. The lowest possible detection threshold is 1 CFU ml-1 while growing each bacterium separately for optimally 6 hours. Influence of additives to the real time PCR efficiency is diverse: bovine serum albumin acts as an inhibitor that reduces DNA amplification efficiency, while ethylene glycol (0,2% - 0,6%) acts as an enhancer that increases DNA amplification efficiency and thus detection sensitivity.
