Investigation of some infectious and genetic factors involved in cervical cancer

dc:descriptionCervical cancer is the second most common cancer in women worldwide and in Vietnam. The causal role of Human papillomavirus (HPV) in cervical cancer has been firmly established. However, there have been only a few studies of HPV infection of the cervix in Vietnam, and in cervical cancer in particular. This project was conducted to investigate infectious and genetic factors that may be involved in cervical cancer in Vietnam. The study comprises two parts: the first part examines the relationship between HPV and Chlamydia trachomatis infections and cervical cancer, and the second part focuses on the impact of HPV infection on microRNA expression. The aims of the first part were to assess prevalence of HPV and its subtypes, C.trachomatis infection and their relationship in cancer of the cervix. The investigation was performed on two populations: Vietnamese (cancer and non-cancer groups), Australian (normal cytology and abnormal cytology groups). The results reveal that (i) both HPV, especially high – risk types, and C.trachomatis are significantly more prevalent in cervical cancer and in cytologically abnormal cervix; (ii) C.trachomatis is a cofactor with HPV and is associated with high-risk HPV types in cervical cancer; (iii) 75% non-cancer women in this Vietnam cohort were infected with high-risk HPVs; (iv) Most of HPV-16 positive Vietnamese samples harbor E6 East-Asia variant whose oncogenicity is potentially greater than the prototype (Europe variant). Together these findings explain in part high rate of cancer of the cervix in Vietnam. The main aim of the second part was to investigate miRNA expression in HPV-16 E6/E7 transfected keratinocytes, in HeLa, SiHa, and C33A cell lines in order to ascertain whether or not HPV and different HPV types affect cellular miRNA expression. These experiments revealed overexpression of some miRNA oncogenes in E6/E7 containing keratinocytes together with underexpression of some miRNA tumor suppressors in (HPV positive) HeLa and SiHa compared with (HPV negative) C33A suggesting that regulation of miRNA expression is an addtional pathway for HPV-induced carcinogenesis.
