Conjugation assays determined that the bacteria mate in a cell density dependent manner, known as quorum-sensing. Analysis of transfer frequencies using a transfer-constitutive mutant of pTiC58 showed that quorum sensing requires both the opine induction and autoinduction systems. Analysis of lacZ fusions determined that quorum-sensing results from TraR and AAI activating transfer gene expression only at high cell densities. The bacteria measure their density by sensing the concentration of AAI in the growth medium.
