High-Resolution Nuclear Magnetic Resonance Microspectroscopy
Abstract
dc:descriptionThis thesis presents a number of improvements and additions, both theoretical and experimental, to NMR microspectroscopy that expand considerably its practical application to studying mass-limited samples. Microcoils have been designed for higher magnetic fields (500 MHz) and also now incorporate a lock channel for longer two-dimensional (2D) experiments. Reference deconvolution and wavelet noise reduction have been used to address lineshape distortion and low signal-to-noise found in NMR microspectroscopy spectra. Double quantum filtered correlated spectroscopy (DQF-COSY) allows signals from selected components in the sample to be isolated. Inclusion of gradients in the microcoil probe greatly enhanced DQF-COSY experiments and enabled a host of other selective editing sequences to be run. Finally experimental measurements and theoretical predictions were shown regarding the effects of restricted diffusion in microcoils. Diffusion-ordered spectroscopy (DOSY) experiments, which allow spectral simplification arising from differences in diffusion coefficient, coupled with conventional chemical shift and scalar coupling differences, were shown for the first time for mass-limited samples.
