Initiation of translation of the small RNA of the cowpea strain of tobacco mosaic virus
Abstract
dc:descriptionSeveral features of the initiation of translation of CcTMV-SRNA were investigated. First, the site of initiation of translation was found to be the first AUG in the 5$\sp\prime$-terminal sequence m$\sp7$GpppGUAUUUGAUGAUGGCA ... despite the fact that this AUG codon is not found within a favorable PuXXAUGG sequence. Second,analysis of the site of initiation of translation of mutant tCcTMV-SRNAs showed that the region of the tandem AUG's must contain some feature which directs the ribosome to initiate translation within that region and that the tandem AUGs themselves may be important for the actual selection of the correct translation initiator AUG codon. Third, investigation of the efficiency of the initiation of translation of the mutant tCcTMV-SRNAs showed that little if any relationship exists between the translational efficiency of the mutant tCcTMV-SRNAs and the agreement, of the sequences surrounding their translation initiation codons, with the PuXXAUGG consensus sequence. Finally, investigation of the translation of the mutant tCcTMV-SRNAs in the presence of $\sp{35}$SMet-tRNA$\sb{\rm i}\sp{\rm met}$ and $\sp{35}$S-Met-tRNA$\sb{\rm m}\sp{\rm met}$ produced unexpected results consistent with the miscoding of AAG codons to incorporate methionine under the conditions used.
