High-throughput profiling of viral class I fusion proteins via mammalian antigen display

Viral infections, particularly epidemics and pandemics, pose a significant threat to global health, causing substantial morbidity, mortality, and socioeconomic disruption. Moreover, viruses can cause chronic disease long after initial infection. While modern technologies enable early outbreak detection, mitigating the burden of future outbreaks requires a deep understanding of infection mechanisms, rapid characterization of viral antigens responsible for host cell entry, identification and profiling of vaccine candidates, and elucidation of how viral evolution enables escape from prior immunity and clinical therapeutics. Here I will review our current understanding of viral entry mechanisms, and the technologies that are commonly used to uncover how viruses infect their hosts, evade immunity, and enable rapid characterization of antigens for future pandemic preparedness.
