Untersuchungen auf Sekundärstoffproduktion und physiologische Charakterisierung von marinen heterotrophen Bakterien aus dem deutschen Wattenmeer
Abstract
dc:description.abstractWithin the investigation of novel marine natural products from previously uncultered bacteria, over 90 isolates from the german Wadden Sea were screened for their potential for secondary metabolite production. 16 S rRNA gene analysis revealed, that 56 strains affiliated with Gram positive bacteria and 36 strains with the Roseobacter-clade (alpha-Proteobacteria). To avoid the problems of culture dependent screening procedures, a PCR-based approach was applied, in which bacteria were screened for polyketide synthase-, FADH2-halogenase- and non-ribosomal peptide synthetase genes. Additionally agar diffusion assays were applied to investigate the antagonistic activity of the strains, and some bacteria were tested for acylated homoserine lactones (AHL) production. Three isolates were additionally screened for synthesis of volatile metabolites, the corresponding compounds were isolated and their structure elucidated.
