Characterization of the Developmentally Regulated Osteoclast Support Potential of In Vitro Medullary Adipocytes

dc:descriptionIn addition to its widely characterized role in lipid storage and energy metabolism, adipose tissue also has an important role as an endocrine organ. Obesity can alter normal systemic physiologic processes and has been linked to diseases such as cardiovascular disease, diabetes, and hypertension. Most studies have used extramedullary adipose tissue as the primary target for adipose characterization. A less-characterized adipose tissue source is the marrow derived medullary adipose tissue. The medullary adipose tissue depot can increase with age and this increase is associated with a decrease in total bone mass and bone mineral density in both mouse and human subjects. Using a well characterized mesenchymal stem cell (MSC)-derived in vitro medullary adipocyte model, the potential for medullary adipocytes to regulate osteoclast development was tested. Factors important in osteoclast development including RANKL, M-CSF and OPG were expressed by in vitro medullary adipocytes. Furthermore, the expression of these important osteoclast mediators was only observed in a specific population of previously unidentified medullary adipocytes at a particular stage of differentiation. Thus, in vitro medullary adipocytes express osteoclast mediators at a restricted developmental stage within the adipocyte lineage that may play a role in bone remodeling.
