A BIOMARKER ENRICHMENT PHASE II TRIAL OF 177LU-DOTATATE IN METASTATIC NASOPHARYNGEAL CANCER WITH A SAFETY RUN-IN
Abstract
dc:description.abstractNasopharyngeal cancer (NPC) is endemic in Southeast Asia. There is a lack of effective targeted treatments for NPC. In this thesis, we investigate the prevalence and prognostic significance of somatostatin receptor 2 (SSTR2) expression in an international study comprising multiple clinical cohorts of 402 primary, locally recurrent and metastatic NPCs. We found significant correlation of SSTR expression with increased rates of survival and report in vivo uptake of the SSTR2-binding 68Ga-DOTA-peptide radioconjugate in PET-CT scanning in a clinical trial of NPC patients (NCT03670342). We also found that SSTR2 expression is induced by the Epstein–Barr virus (EBV) latent membrane protein 1 (LMP1) via the NF-κB pathway and SSTR2 expression can be seen in vitro and in vivo in certain other EBV related cancers (NCT05581550). We also report the initial results of our phase 2 therapeutic trial (NCT05198479) to study the efficacy of targeting SSTR2 with 177Lu-DOTATATE in metastatic NPC.
