Our data suggest that the PPARalpha/NCoR interaction is physiologically relevant, and can produce a discernable phenotype when the magnitude of the interaction is altered by a naturally occurring variation. Our detailed mechanistic study of the PPARalpha V227A variant allows for the design of future human studies to identify other benefits and risks associated with this mutation. Furthermore, the identification and characterization of isoflavones, and their parent botanicals, with different PPARalpha/gamma potencies suggest their value in the management of the epidemic of diabetes, dyslipidemia and the metabolic syndrome.
