GENETIC REQUIREMENTS OF DANGEROUS MIX AUTOIMMUNITY IN ARABIDOPSIS THALIANA
Abstract
dc:description.abstractAutoimmunity is often found in hybrid plants due to uncontrolled immune responses without obvious pathogenic attacks. Due to the characteristic strong phenotype of hybrid necrosis, it has been successful in identifying DANGEROUS MIX (DM) genes in Arabidopsis thaliana, consisting of two incompatible immune components originating from each parent. In my thesis, I utilize the autoimmunity-causing DM pairs to study the mechanism of immune signaling activation and downstream immune signaling pathways. To facilitate genetic requirement studies, I first optimized a versatile CRISPR tool to edit core immune components in A. thaliana. The autoimmunity mechanism of DM6-DM7 and DM10-DM11 was pursued by researching on the genetic requirements and biochemical properties. Knowledge gained from the study of genetic requirements of the two autoimmune cases will help us advance our understanding on the activation mechanism of immune receptor, signal transduction and cell death execution in the plant immune system.
