Osteogenic and chondrogenic differentiation of rat bone marrow stromal cells in 3D culture with novel PCL-TCP scaffolds

The aim of this project was to investigate the efficacy of osteogenic and chondrogenic differentiation of rat bone marrow stromal cells (BMSC) in 3D PCL-TCP scaffolds. PCL-TCP scaffolds fabricated by fused deposition modeling (FDM), are honeycombed, highly reproducible bioresorbable 3D structures with a completely interconnected pore network and custom-tailored porosity and pore size. After expansion in vitro, BMSCs were enzymatically lifted and then seeded with fibrin glue as cell suspensions into PCL-TCP scaffolds. The cell-scaffold constructs were cultured for 3 weeks in vitro. Cell viability, morphology, and distribution were examined by phase-contrast microscope, confocal microscope, MTT assay, and SEM. Osteogenic differentiation was substantiated by Von Kossa staining, heightened expression of ALP and osteocalcin; chondrogenic differentiation was confirmed by alcian blue staining and toluidine blue staining. In conclusion, it was demonstrated that PCL-TCP scaffolds with fibrin glue facilitate proliferation, anchorage, motility, and differentiation of both specialized cells and BMSCs.
