Susceptibility Testing of Pathogenic Fungi With a Radiometric System
Abstract
dc:description.abstractA radiomethric method was used to determine the susceptibility of clinical isolates of dimorphic fungi (Histoplasma capsulatum, Blastomyces dermatitidis, Sporothrix schenckii) to selected antifungal agents. Antifungal agents evaluated were Amphotericin B, 5-fluorocytosine, Nystatin, Ketoconazole, Miconazole, Terconazole, and Itraconazole. The instrument used to measure amount of radioactive carbon dioxide generated by fungal metabolism of ¹⁴C-glucose in presence of antifungal agents was the Bactex 460. Mycelial cultures were grown on SAB-DEX agar at 25°C and converted to yeast phase in a chemically defined, liquid medium at 37°C. Yeast inoculum was adjusted to 5x10⁴ cells. Results indicate that the radiometric method provides a reproducible, rapid, and quantitative measurement of the inhibitory effects of antifungal agents on yeast growth in one-third the time required with conventional susceptibility test methods. This radiometric method has potential for routine laboratory use in evaluating antibiotic susceptibility of fungi
