Growth and Characterization of Cell Cultures of Rainbow Trout

Gonad and spleen primary cultures of rainbow trout (Salmo gairdneri Richardson) were initiated and compared to the rainbow trout gonad transformed cell line RTG-2. The RTG-2 cell line grew faster than the gonad primary cultures. The chromosome number of the gonad primary culture was 2n=56 with 104 chromosome arms. The RTG-2 cell line chromosome number ranged from 2n=48 to 2n=59. Giemsa and reverse banding of the chromosomes were unsuccessful. Protein differences were observed between the gonad primary culture, spleen primary cultures, and the RTG-2 cell line as detected by molecular weight determination using SDS-polyacrylamide gel electrophoresis.
