The Role of the Parotid Salivary Glands in the Regulation of Insulin Secretion
Abstract
dc:description.abstract<p>Stress of handling, partial or complete sialoadenectomy, and the administration of parotid hormone (PH), sodium fluoride (NaF), or guanidino-succinic acid (GSA) were examined for effects on fasting plasma glucose and insulin or effects on intravenous glucose tolerance in male Sprague-Dawley rats. Basal glucose and insulin were substantially elevated as a result of the stress of translocation in large animals, but were not altered in small animals. A rise in plasma insulin occured in control animals 20 minutes following anesthesia and the removal of a blood sample by cardiac puncture but not in animals that had been handled twice daily for one week prior to the experiment.</p> <p>Salivary gland removal did not alter basal insulin or glucose or short (10-minute) glucose tolerance (SGT). Results of long (60-minute) glucose tolerance tests (LGTs), however, suggest that parotidectomized animals secrete more insulin in response to a glucose challenge.</p> <p>PH and GSA administration did not alter glucose tolerance or basal glucose and insulin. Sodium fluoride pretreatment did not change basal values or SGT results, but did increase post-stimulus glucose levels in LGTs. Also, the data from LGTs with NaF pretreatment suggest that NaF increases or prolongs the insulin response to glucose infusion.</p>
